New Insights on Weight-Loss Drugs and Cancer Research
Recent studies are exploring the impact of GLP-1 medications, which are designed to treat obesity and diabetes, on cancer risk and outcomes. The American Association for Cancer Research (AACR) released its 2026 Cancer Progress Report, highlighting obesity as a significant risk factor for various types of cancer.
Medications like semaglutides (found in popular drugs such as Ozempic and Wegovy) and tirzepatides (like Mounjaro and Zepbound) have shown remarkable results in weight loss and improving metabolic health. This has caught the attention of researchers focused on evaluating their potential effects on cancer.
While the AACR report referred to some studies suggesting that GLP-1 medications might lower the risk of certain cancers, it stressed that the evidence is not clear-cut and that further investigation is necessary. According to the report, large-scale clinical trials specifically aimed at assessing cancer outcomes related to GLP-1 receptor agonists are still needed.
Dr. Sai Yendamuri, a thoracic surgeon at Roswell Park Comprehensive Cancer Center, pointed out that obesity is a significant cause of cancer, ranking second only to smoking in terms of risk factors, while other issues like alcohol consumption and nutrition also contribute to obesity. He posed an important question: “If obesity is linked to cancer, could a drug that reduces obesity positively affect cancer risks?”
Dr. Yendamuri indicated that there is substantial evidence supporting the idea that weight loss might reduce cancer risk. He referenced research showing that patients who underwent metabolic surgery for obesity experienced a decrease in cancer rates afterward.
Many studies examining patients on GLP-1 receptor agonists have reported lower cancer incidence and better survival rates compared to those not on these medications. However, it’s important to note that some studies found no link between these medications and cancer rates, while a few indicated an increased risk for specific cancers, such as thyroid cancer.
The variability in results might be due to the fact that many of these studies are retrospective, meaning they analyze existing data rather than conducting new trials. This can lead to different conclusions based on various factors.
Dr. Yendamuri’s research has consistently shown that obesity raises the risk of lung cancer. Moreover, he mentioned that defining and measuring obesity accurately is vital, as reliance on Body Mass Index (BMI) alone may not capture the complete picture of body composition.
He shared that in lung cancer surgeries, patients on GLP-1 medications tend to have lower recurrence rates, suggesting that these drugs might help the body’s immune system respond better to cancer. He noted, however, that the effect of GLP-1 medications may vary based on cancer type, with immune response playing a critical role in some cancers like melanoma.
Dr. Yendamuri acknowledged that part of the potential cancer benefit from GLP-1 medications likely stems from weight loss, but not every patient will see significant weight changes while on these drugs. Future studies should explore whether the benefits of these medications depend on actual weight loss.
Overall, the growing understanding of the relationship between lifestyle factors, such as weight management and exercise, and cancer prevention reinforces the need to consider integrated approaches to cancer treatment. Dr. Yendamuri advocates for ongoing support for the National Cancer Institute to continue advancing cancer research and discovery.
