While the APOE4 variant is linked to a higher risk of Alzheimer’s, the APOE2 variant appears to be beneficial, associated with a lower risk of cognitive decline.

Researchers from the Buck Institute for Research on Aging looked at human brain cells derived from stem cells to understand how APOE2 provides this protective effect. They discovered that APOE2 boosts the ability of neurons to repair DNA damage and withstand aging, while neurons with the APOE4 variant showed more signs of aging and dysfunction.

Follow-up research in mice supported these findings. Lisa M. Ellerby, the lead researcher and a professor at the Buck Institute, explained that APOE2 helps neurons manage DNA repair better, contributing to its link with longevity and a reduced risk of Alzheimer’s.

The study also revealed that introducing the APOE2 protein to neurons with the APOE4 variant reduced DNA damage caused by stressors like radiation. This suggests that future therapies might aim to mimic APOE2’s protective effects or enhance DNA repair mechanisms in the brain, especially for those at higher risk due to the APOE4 gene.

Christopher Weber from the Alzheimer’s Association said this research is exciting as it highlights a new function of the APOE gene beyond its known role in cholesterol transport. He noted that understanding how the brain maintains its health as it ages could lead to new therapies for those carrying high-risk genes.

Additionally, researchers emphasized that while their findings are promising, they were based on lab studies and further investigation in living patients is required to confirm these effects. They also cautioned against making lifestyle changes solely based on this study.

Researchers recommend maintaining overall brain health through regular exercise, quality sleep, good heart health, and avoiding harmful substances like tobacco. These strategies can be beneficial for everyone, regardless of their APOE variant.

Study Reveals Longevity Gene May Protect Brain from Aging A recent study indicates that a gene known as APOE may play a crucial role in protecting the brain from aging-related issues, including Alzheimer’s disease.
