New Drug Shows Promise in Slowing Aging Process While Treating Lung Disease
Researchers have made an exciting discovery regarding a new drug designed to combat a serious lung condition, suggesting that it may also help slow the aging process.
In a small clinical trial, patients who were given rentosertib, a drug developed by Insilico Medicine in Massachusetts, displayed changes in their blood protein levels that indicated a more youthful biological age. According to the findings, participants appeared to be around three to four years younger after treatment, with one individual showing a change equivalent to six years.
Alex Zhavoronkov, PhD, the first author of the study and co-CEO of Insilico Medicine, explained, “This shows a promising trend in age reversal alongside treating the disease, which is a valuable bonus.” He added that the research utilized validated methods to gauge biological age, though the implications of these findings are still being explored.
The study results were shared in the journal Nature Biotechnology and presented at a conference at Sorbonne University in Paris. While the results are promising, researchers warn that this does not confirm that the drug can reverse aging or increase lifespan. “It’s early days,” Zhavoronkov noted, emphasizing that rentosertib is still under investigation.
Insights into Rentosertib
Rentosertib is an experimental drug aimed at treating idiopathic pulmonary fibrosis (IPF), a serious condition that leads to lung scarring. The drug was developed using artificial intelligence, and unexpected benefits were observed, with patients showing a younger biological age after treatment.
In this study, six different tests assessed biological aging, and results consistently indicated that those taking rentosertib were biologically younger than those receiving a placebo. The most significant effects were noted four weeks into the treatment, particularly in patients taking a dose of 30 mg twice daily.
When comparing these results to data from over 55,000 individuals in a UK health study, scientists found that rentosertib altered certain blood proteins in a way that contrasts the typical patterns seen with aging. Additionally, it indicated changes at the cellular level that adjust how cells operate and respond to stress.
Zhavoronkov remarked, “Everyone agrees that living longer should be a fundamental human right for all.”
This study involved several prestigious institutions, including Harvard Medical School and Stanford University, working collaboratively with Insilico Medicine.
Study Limitations
Despite these encouraging results, the study had a few limitations. Zhavoronkov pointed out that the findings are preliminary and exploratory, with only 42 patients analyzed over a 12-week period. All participants had IPF, which may have affected the proteins related to measuring biological aging. This means that the observed changes could be tied to improvements in the patients’ lung condition.
Future trials should involve larger groups and include healthy older adults to better understand whether rentosertib does indeed have anti-aging benefits and contributes to long-term health.
Potential Side Effects
Dr. Carol Ann Satler, a senior vice president at Insilico Medicine, reported that the side effects associated with rentosertib are manageable. Some common side effects noted include mild diarrhea, lower potassium levels in the blood, and temporary increases in liver enzymes. She explained that once treatment stops, any benefits will likely return to previous levels.
Looking ahead, the next phase of testing will involve adjusting dosages based on side effects, allowing patients to stay on the medication longer. Rentosertib has now entered a year-long phase 3 trial involving 300 patients in China.
Although more research is needed, Zhavoronkov is hopeful, stating, “These initial findings reassure us in our pursuit of drugs that aim to tackle both age-related diseases and the aging process itself.”
